OK, so never having handled nucleic acids in anger before (shut up at the back), today I am actually starting my reverse-transcription PCR optimisation. Cue much messing about in UV cabinets and miscalculating dilutions of mastermix.
Have already found out that I don't have enough kit to fully optimise the reaction for all of my primer pairs, let alone do the experiment that I bought the kit for in the first place. Replicates, so many replicates... because 5 primer pairs are more likely to throw up a working scenario than one. Biology, such a frikkin fuzzy science.
